If you think about it, Chinese folklore has millennia of content, refined for centuries, to inspire movie makers. All they need is some computer graphics, decent acting and some experience and they are set to build fantastical worlds that the Western World hasn't even dreamed of. This film is part of a sort of thematic series, with another Yinyang Master movie from 2020 having absolutely no connection to this one, but set in the same world of Chinese mythology. And it was good!Now, the scenes were not really polished, the actors and their dialogues not to the standards that we've come to expect from Hollywood productions, but that's as much a good thing as a bad thing. The CGI was a bit clumsy only with the "Red Devil" character and maybe the final fight, the rest being pretty spot on. But what I liked more was the story, which had complex characters, hero journeys, a big baddie and tons of fun fights that kind of made sense.Bottom line: this is one of the most entertaining and accessible Chinese films I've seen in a long time and, what's best, it felt natural, not an attempt to clone a particular Western genre. It felt authentic as well as interesting. Keep up the good work, guys!
Special Episode Part 3 is the third of five extra episodes unlocked after completing the final main story episode of Wario: Master of Disguise. It takes place in the Ancient Waterworks. The objective, as is the case with the other special episodes, is to obtain three treasures in order within a time limit.
The first step conducts quality assessment and generation of a report. Next, sequences are mapped to a reference genome and the methylation information is extracted. Based on several quality statistics and thresholds the generated results are filtered and aggregated into a final output table. In the next step, reads covering all CpGs in a target region are used to calculate methylation-pattern statistics, which are subsequently compared between samples. The last step creates the final output table, graphical representations of the results, and reports basic statistics generated during the analysis workflow.
The output creation module aggregates the generated methylation information of all individual targets for all samples into a final table (see Fig 3). Several filtering steps are applied to remove false methylation calls (see section Using TABSAT): first low coverage methylation calls are removed from the output using a cutoff filter that can be set as a parameter. In addition, TABSAT determines the average coverage for each CpG per target/sample using the methylated and unmethylated coverage information. Next, it calculates a mean from these averages for each sample/target combination, which is used to remove every methylation call where the total coverage of a CpG is lower than the calculated mean coverage minus one standard deviation. As we use an amplicon sequencing approach, we expect a uniform coverage for each sample/target. By taking the average coverage of all CpGs within one sample/target and comparing it to the sum of each CpG we ensure to remove only extreme outliers. In addition, all CpGs that are not on the user specified strand are filtered out.
We have developed a novel tool for the analysis of targeted bisulfite sequencing, which is especially equipped to handle, in addition to Illumina data, sequences generated on Ion Torrent systems. To date, several tools exist for the analysis of bisulfite data (reviewed here ), amplicons from bisulfite flowgram sequencing (Amplikyzer ) and locus-specific analysis of 5-methylcytosine (BiQ Analyzer HiMod ). However, none of these tools is specifically tailored for the analysis of Ion Torrent sequencing data and provides a one-stop solution from raw sequencing data to final results. Furthermore, the Ion Torrent PGM software platform currently does not support the analysis of bisulfite sequencing data. TABSAT comprises an analysis pipeline containing quality control, alignment, methylation calling, and output generation. In order to select the best mapping software for our purpose, we have evaluated several bisulfite analysis programs, such as Bismark , BS-Seeker2 , and BSMAP . All programs produced similar or poorer mapping results, and offer different downstream analysis capabilities. Preliminary analysis with one input file using default parameters resulted in around 45%, 42%, and 20% of mapped reads for Bismark, BsSeeker2, and BSMap, respectively (see _tools). Based on the availability of the source code, the possibility to integrate a different mapping program, and the positive reviews, we decided to include Bismark in our workflow to handle mapping of sequencing reads and methylation calling.

The detailed protocol is available here but essentially the aim of this step is to cover Ion Sphere Particles (ISPs) with a single template by emulsion PCR. The primers on the ISPs bind to the adapters added in the library prep. The sequence bound on the ISP undergoes PCR and replicates all over the ISP. It is important to get the input concentration correct so that there are enough amplicons to provide sufficient data, but also that the concentration is not too high, as if too high, more than one amplicon can end up on the ISP (to give a polyclonal ISP) which cannot be used and will be discarded from the final data set.
This is where we finally load the pooled libraries (which have been annealed to the ISPs) onto the P1 chip with control ISPs also. We anneal the sequencing primer to the eISPs and then the aim is to get the ISP into the 148,155,732 wells on the chip
Next we want to make sure that the ISPs were prepared and enriched properly. We can see that in this table. In this example 95% of the wells have ISPs which is good (the best we usually achieve), nearly all of those are library ISPs. The polyclonal ISPs I described earlier are disregarded, as are those of low quality. This then gives our final number of reads.
The torrent comes ready for ITX, ATX, and EATX boards with only mATX or server format boards needing the standoffs adjusted. The center standoff is a pin that helps align and hold the board in place while you fasten it down.
Only two Elden Ring characters mention Torrent by name: Melina and Ranni. Both women seem to have known Torrent's former master, well enough for them to be entrusted with the Spectral Steed Whistle and Spirit Calling Bell to pass onto the Tarnished. Yet Torrent's former master is never mentioned by name or expanded upon beyond the the two items that are to be given to Torrent's next master. It's also interesting to note that Melina and Ranni are spirits like Torrent, with Melina saying she is bodyless and Ranni admitting that she cast aside her body willingly. This further adds to the mystery of Torrents former master, leaving players to wonder why they'd choose characters who are spirits to pass on their spectral steed, or if the reason they chose Ranni and Melina is not related to their lack of bodies at all.
As financial reports hint at Elden Ring DLC, there's hope that some of the lingering questions about the Lands Between and its residents will be answered. There's too much set-up in lore for Torrent and his former master for no more information about either of them to be given. Players deserve to know more about their spectral steed, who is first presented as a character in his own right, and if they may have encountered or fought Torrent's former master in Elden Ring.
The master gave me the foundations and a good theoretical framework and knowledge to successfully work in the development field, being able to understand how the international system works, giving me the capacity to analyse public policies across various sectors, the role of the different players in the international field, how policy making takes place, how policies are delivered and evaluated, etc. It also widened my mind and switched my criticism on, and made me realise about the importance of questioning things. Since the master was quite demanding in terms of delivering outcomes I also learned to work fast, to become more efficient, to better work in groups and specially to work in a multicultural environment, something that later on proved to be very important
Yes and no. What I pictured after completing the master's was me working for an international organisation. And I really had the chance to do so. Last April I was accepted to work in a multilateral institution in Budapest, but I finally declined the position for personal reasons. I have now become an entrepreneur and freelance consultant, and to be honest, I am quite happy about it, but this was not something I was expecting in the beginning. As somebody told me many years ago: 'you know where your career path starts but not where it ends'.
